ABSTRACT

Objective@#To investigate the effects of maternal exposure to ambient fine particles (PM2.5) in Fuzhou during pregnancy on immune responses to ovalbumin (OVA) in neonatal rats and the possible mechanisms.@*Methods@#Pregnant Sprague-Dawley rats were randomly assigned into four groups (ten in each): filtered air (FA) plus normal saline (NS), airborne PM2.5 plus NS (PM2.5-NS), FA plus OVA (FA-OVA) and PM2.5 plus OVA (PM2.5-OVA) groups. Pregnant dams in the PM2.5 exposure groups were placed in a PM2.5 exposure chamber in which the PM2.5 concentration was equal to the ambient air from the beginning of gestation till delivery, whereas the other dams inhaling air without particulate matters were put into a clean chamber. OVA sensitization was conducted through intraperitoneal injection of OVA at 50 μg per dam at 4 and 9 days of gestation, followed by inhalation of atomized 1% OVA for 30 min at 18, 19 and 20 days of gestation. Dams without OVA sensitization were given NS in the same way. Levels of interleukin (IL)-4, IL-5 and interferon-γ (IFN-γ) in neonatal rats' plasma were measured by enzyme-linked immunosorbent assay just after birth. Protein levels of transcription factors GATA-3 and T-bet in lung were analyzed by Western-blotting. Changes in microRNA(miR)-146a and miR-146b in spleen were detected by real-time polymerase chain reaction. Histological changes in lung were observed under light microscope. One-way analysis of variance and LSD test were used as statistical methods.@*Results@#(1) IL-4 level in plasma was significantly increased in PM2.5-NS [(18.56±7.04) ng/L], FA-OVA [(34.04±7.06) ng/L] and PM2.5-OVA groups [(45.67±8.18) ng/L] as compared with that in FA-NS group [(10.51±2.88) ng/L], and the level of IL-4 in PM2.5-OVA group was higher than that in PM2.5-NS and FA-OVA groups (F=54.667, P<0.001). Significantly increased IL-5 level in plasma was found in FA-OVA and PM2.5-OVA groups as compared with that in FA-NS group (F=6.253, P=0.023). Among the four groups, FA-OVA group showed significantly increased IFN-γ level in plasma (F=28.604, P<0.001). (2) GATA-3 level in lung tissues was significantly increased in PM2.5-NS (31.09±3.54), FA-OVA (35.24±5.00) and PM2.5-OVA groups (47.81±3.63) as compared with that in FA-NS group (24.19±3.12), and higher in PM2.5-OVA group than in PM2.5-NS and FA-OVA groups (F=96.581, P<0.001). T-bet level was significantly lower in FA-OVA and PM2.5-OVA groups than in FA-NS group. Moreover, PM2.5-OVA group showed decreased T-bet level as compared with that in PM2.5-NS and FA-OVA groups (F=30.852, P<0.001). (3) Expression of miR-146a in spleen was significantly enhanced in PM2.5-NS (1.72±0.27), FA-OVA (1.56±0.37) and PM2.5-OVA groups (3.06±0.52) than in FA-NS group (1.05±0.25). Moreover, PM2.5-OVA group showed enhanced expression of miR-146a as compared with that in PM2.5-NS and FA-OVA groups (F=42.276, P<0.001). Changes in the expressions of miR-146b were similar to those in miR-146a (F=28.776, P<0.001). (4) Stenosis or disappearance of alveolar spaces, accompanied with infiltration of inflammatory cells in interstitial substance and congestion in alveolar septum, was seen in FA-OVA and PM2.5-OVA groups and conditions in the latter group were more severe.@*Conclusions@#Intrauterine exposure to ambient PM2.5 negatively affects fetal lung development and immunological function in rats, especially when the dams are sensitized with OVA during pregnancy.

ABSTRACT

High incidence of postoperative abdominal adhesions after gynecological laparoscopy, which may induce a line of serious complications such as chronic pelvic soreness, intestinal obstruction, infertility, etc, has been clinically encountered with the major problems unsolved thus far. By further upgrading the merits of gynecological laparoscopy, including the small operative scope, mild injury and fine manipulation, coupled with the application of anti-adhesion therapies, reduced post-operative adhesions and improved clinical outcomes can be expected. Research has been actively persisted on this area in recent years to achieve new progress, especially in the development of anti-adhesion materials, as well as their clinical application and evaluation of the effects, which is overviewed in this review.

ABSTRACT

Objective To investigate the relationship between maternal exposure to airborne inhalable particulate matters (PM10) in the first trimester of pregnancy and the risk of fetal congenital deformity. Methods Relationship between exposure to airborne PM10 during the 1st,2nd,3rd and 1-3 months of gestation and the risk of fetal anomalies in 203 pregnant women with deformed fetuses, which paralleled to normal pregnant women with discrepancy of conception date <30 days (control),were retrospectively analyzed by a case-control study from May 14,2007 to April 30,2012 in Fujian Medical University and Fuzhou General Hospital.Multivariate Logistic regression adjusted for potential confounders including maternal age, gravidity and parity was performed for data analysis. Results According to the inclusion and exclusion criteria,178 pregnant women with deformed fetuses and 356 controlled cases were enrolled. The average levels of PM10 exposed in pregnant women Withfetal cardiovascular anomalies during 1-3 months of gestation were significantly higher than in the controls [(73.80±11.55) μg/m3 vs (70.49±10.83) μg/m3] (t=2.066,P=0.040),but PM10 exposure in the 1st,2nd and the 3rd month of pregnancy were comparable between the observed and control groups [(74.00±17.34) μg/m3 vs (71.70±15.39) μg/m3,t=0.992,P=0.322;(75.15±16.80) μg/m3 vs (71.38±15.66) μg/m3,t=1.625,P=0.106; (70.28±15.84) μg/m3 vs (69.41± 15.84) μg/m3,t=0.383,P=0.702].There were no significant differences of PM10 exposure levels when compared between facial-cervical anomalies, neurologic anomalies and total anomalies and their corresponding controls (P>0.05).Each increased quartile of the PM10 exposure levels was associated with an elevated risk of fetal cardiovascular anomalies by 1.218 folds (OR =2.218,95 % CI:1.232-3.994,P=0.008).Paired multivariate Logistic regression analysis revealed,a positive correlation between PM10 levels at 1-3 months of gestation and fetal cardiovascular anomalies (OR =1.106,95%CI:1.035-1.183,P =0.003).Conclusions Maternal exposure to PM10 in the first trimester of pregnancy possibly exerts negative effects on fetal cardiovascular anomalies.

ABSTRACT

Objective To investigate subacute exposure of airborne particulate matter (PM) on pregnancy and fetal development in female mice. Methods Forty female and forty male ICR adult mice group (A), small (B) , middle (C) , large (D) or overdose (E) PM challenge groups (n = 8 - 11), and were administered with 30 μl of phosphate buffered solution (A) or resuspended standard PM SRM 1649a at 0.09 (B), 0.52 (C), 1.85 (D) or 69.2 (E) μg/μl, once per trid from d 0 till d 19 of pregnancy via instillation onto the base of the tongue. Fetal mice were harvested by cesarean section at the time when spontaneous delivery occurred. Body weight of the pregnant mice, gestational days, intrauterine survival and growth, hepatic and pneumonic histopathological changes of the fetal mice were investigated. Lung/body and liver/body weight ratios were calculated. Expressions of mRNA and protein of CYP1A1 in the fetal lung and CYP1 A2 in the fetal liver were assayed. Results (1) All of the pregnant mice survived pregnancy throughout the entire experiment. Body weight of the pregnant mice was not significantly different among all the groups at gestational d 1 and 7 (P > 0.05), but significantly lower in group E [(41.8 ± 5.8) and (48.9 ± 8.9) g] than in group A [(45.9 ± 1.8) and (56.2 ± 4.9) g] at gestational d 14 and 18 (P <0.05). The gestational days were significantly decreased in group E [(19.3 ± 1.3) d] when compared with group A [(20.5 ± 0.7) d; P < 0.05] and were not significantly different among groups A, B, C and D (P > 0.05). Lung/body and liver/body weight ratios of the fetal mice were significantly increased in group E [(1.21 ±0.18) and (4.68 ±0.21)%] as compared with groups A, B, C and D (P<0.05). (2)Mortality rates of the fetuses were significantly higher in group E (23.0%) than in groups A (0.8%), B (0.9%), C (1.7%) and D (3.7%) (P < 0.05), but were not significantly different among groups A,B, C and D (P > 0.05) despite of an increasing tendency. (3) Pathological changes in the liver and lung of the fetuses were conspicuous in group E. The fetal liver injury was histopathologically evidenced by deranged tissue structure, degenerated parenchyma of hepatic cells, and mildly stained cytoplasm. Adipose degeneration was represented by clear-boundary intracytoplasmic vacuoles in most of the liver cells, and cell pyknosis with heavily stained cytoplasm was observed in some of the liver cells. Inflammatory cell infiltration and focal necrosis were occasionally found in the hepatic tissue. The fetal lung exhibited bronchiole with narrow lumina, vascular engorgement in the submucosal layer, interstitial and alveolar edema, thickened alveolar septum, granulocyte and lymphocyte infiltrations within the pulmonary alveoli and around the bronchioles. The above pathological changes were lesser in groups C and D, and were not or least found in groups A and B. (4) Protein expressions of CYP1A1 in the fetal lung and CYP1A2 in the fetal liver were significantly increased in group E (1.20 ± 0.40 and 2.55 ± 0.89) when compared with group A (0.77 ±0.36 and 2.08 ±0.31) (P < 0.05). mRNA expressions of CYP1A1 in the fetal lung were significantly increased in groups C (0.36 ±0.12), D (0.41 ±0.08) and E (0.43 ±0.11) compared with group A (0.21 ±0.10), and significantly increased in groups D and E compared with group B (0.28 ±0.10,P<0.05). mRNA expressions of CYP1 A2 in the fetal liver were significantly increased in groups C (0.37 ±0.13), D (0.36 ±0.14) and E (0.43 ±0.16) compared with group A (0.21 ±0.03), and significantly increased in group E compared with group B (0.24± 0.11, P < 0.05). Conclusions PM elicited embryotoxigenicity and resulted in adverse pregnancy outcomes in mice by intrauterine exposure of overdose PM. The expressions of cancer-related genes CYP1A1 and CYP1A2 were up-regulated in organs after the middle- and large-dose subacute exposure of PM, which may have a potential role on the future development.

ABSTRACT

Objective To investigate the effects of compound radix angelicae sinensis injection on oocyte and segmentation sphere injured by subacute exposure to diesel exhaust particles (DEP) in female mice. Methods Two hundred and ten 21-day-old ICR female mice were randomly divided into 5 groups: the control group (group A), DEP group (group B), DEP+low-dose group (group C),DEP±middle-dose group (group D) and DEP+high-dose group (group E). There were 42 mice in each group. The mice were inoculated with 30 μ1 DEP suspension at 12.0 μg/μl (group B-E) or the same volume of vehicle (PBS, group A) on pharynx posterior wall by sample pipettor beginning at day 21 and repeated every 3 days for 4 times. The mice were sacrificed three days after the last exposure.Compound radix angelicae sinensis injection containing 75 (group C), 150 (group D) and 300 (group E)grams of crude drug, respectively, which was intraperitoneally administered for each mouse daily from the day of the first DEP inoculation till the day before sacrifice, consecutively for 12 days. The general conditions were observed, and the body weight and ovary/body weight ratio were tested. Superoxide dismutase (SOD) activity, malondialdehyde (MDA) and reduced glutathione (GSH) contents in ovarian tissues were assayed. Rates of survival, germinal vesicle breakdown, extrusion of the first polar body and in-vitro fertilization, and quantity of mitochondrial DNA for oocytes were investigated.Ultrastructural changes of oocytes were observed. Results ( 1 ) No significant difference of the body weight was found among all the groups (P>0. 05). The ovary weight, ovary/body weight ratio, ovary SOD and GSH content were significantly decreased in groups B [( 1.5 ± 0. 6) mg, ( 7.2 ± 2. 5) × 10-5 ,(192. 10±23.67) nU/mg prot and (262.40 ± 31.60) nmol/mg prot], and C [( 1.7 ± 0. 2) mg,(8.9±0.6)× 10-5, (198.92±24.27) nU/mg prot and (271.66±14.58) nmol/mg prot] and D [(2. 1±0. 2) mg, (9. 8±1. 1)×10-5, (214. 37±27. 19) nU/mg prot and (285. 93±9. 55) nmol/mg prot] as comparing to groupA [(3. 3±1. 5) mg, (15.4±7.3)×10-5, (292. 30 ± 40. 03) nU/mg prot and (367.98±24.59) nmol/mg prot (P< 0. 05 or P<0.01); and significantly increased in group E [(3. 7±1.1) mg, (18. 7±5. 4)× 10-5, (279. 10±12. 63) nU/mg prot ]and (353. 59±10. 61) nmol/mg prot]comparing to group B (P<0. 01). MDA content was signi-ficantly increased for groups B, C and D [(3. 88±0.35) nmol/mg prot, (3. 62 ± 0. 19) nmol/mg prot and (2. 63 ± 0. 34) nmol/mg prot] comparing to group A [(2. 18±0. 44) nmol/mg prot](P<0. 05 or <0. 01, respectively); and significantly decreased for group D and E (2. 35±0. 37 nmol/mg prot) comparing to group B (P<0. 01). (2) In all observed time points, oocyte survival rate in group B and C, extrusion rate of the first polar body and in-vitro fertilization rate in group B, C and D were significantly lower than in group A (P < 0. 05 or <0. 01), and all those in group E were significantly higher than in group B (P<0. 05). Rates of germinal vesicle breakdown were 100% in all five groups. (3) Logarithmic values of mitochondrial DNA copy numbers in group C, D and E were significantly lower than in group A; whereas significantly higher in group C and D than in group B (P<0.01). (4) Normal appearance for oocytes in group A was seen. In groups B and C, a number of cytoplasmic organelles were dramatically degenerated in part of the oocytes and some necrotic oocytes were seen. Large body of mitochondria in the oocytes swelled and vacuolized in group D, while such changes dincished to a lesser extent and scope in group E. Conclusions Compound radix angelicae sinensis injection exerts a favorable curative and protective effect on oocyte and segmentation sphere injured by subacute DEP exposure in female mice.

ABSTRACT

BACKGROUND: Endogenous opioid peptides are mainly mediated by μ receptor and produce inhibitory effect on breathing and heartbeat, and because of this, fetal distress happens and develops. Therefore, μ receptor mediation is the most important way that endogenous opioid peptides participate in the fetal distress and the pathological process of suffocation. Β-FNA (β-Funaltrexamine, μ-receptor antagonist) and ICI174864(δ-receptor antagonist) are helpful to reduce the happening of fetal distress.OBJECTIVE: To explore the roles of μ and δ receptors in the fetal distress based on the point of view that opioid peptides are involved.DESIGN: Completely randomized allocation and randomized and controlled trial using experimental animals as the subjects.MATERIALS: Twenty healthy white New Zealand purebred rabbits were randomly divided into 5 groups, and among them 16 rabbits were pregnant for 30 days and were put to death with the method of suffocation. The fetal rabbits were taken out immediately after cesarean section, with 6 - 8 fetal rabbits in each den. The fetal rabbits were classified into the following groups: untreated fetal distress group, (fetal distress group, 29 fetal rabbits), normal saline rabbits group(Normal saline group, 25 rabbits), rabbit distress β-FNA group(β-Funaltrexamine ) (FNA group, 28 rabbits), fetal rabbits ICI174864 treatment group(ICI174864 group, 31 rabbits).INTERVENTIONS: Pregnant rabbits in the saline group, FNA group , ICI group were intravenously given a bolus of normal saline or the opiate antagonist β-FNA or ICI174864 and were then asphyxiated. Another 4 rabbits that were pregnant for 30 days were put to death with the cut on the neck, and 28 fetal rabbits were taken out as the control group. After being delivered by cesarean section, all rabbit fetuses in the above five groups were assessed by Apgar scoring for respiration, heartbeat, skin color, muscle tension, and response to stimulation at 1, 5, 10, 15 and 30 minutes of age.MAIN OUTCOME MEASURES: Apgar scoring for respiration, heartbeat, skin color, muscle tension, and response to stimulation at 1, 5, 10, 15 and 30 minutes of age was made.RESULTS: The total Apgar score for respiration, heartbeat, skin color, muscle tension in the control group was the highest (8. 8 ± 1.1 ). And that of FNA group was 6. 8 ± 1.7, obviously higherthan that of the fetal distress group (2.1 ±1.0) and the saline-treated pups(2.5±1. 1) and(t=2.832 and 2. 795, P < 0.01 ). That of group ICI was 4.9 ± 0.7, markedly higher than that of the saline-treated pups and the fetal distress group( t = 2. 232 and 2. 195, P < 0.05) . There was no marked difference between the saline group and the distress group ( P > 0.05 ) and between the FNA group and the control group(P < 0.05) . The total Apgar scores of the ICI group, saline group, and distress group were significantly lower than that of the control group ( t = 2.913,2. 893, P < 0.01 and t = 2. 174, P < 0.05).CONCLUSION: Endogenous opioid peptides participates in the process of the happening and development of fetal rabbit distress that is mediated by opioid peptides receptor, the effect of μ receptor was much more than that of δ receptor. This study provides a basis for the evaluation of fetal distress and intervening at an earlier stage to enhance prognostic function.

ABSTRACT

BACKGROUND: Endogenous opioid peptide is an important medium and regulator that participate in many physical and pathologic processes of the body. Its relationship with fetal encephalopathy has attracted much attraction.OBJECTIVE: To evaluate the role of endogenous opioid peptides (EOP) in fetal distress.DESIGN: A case-control observatory study based on healthy pregnant women.SETTING: Wards of the Department of Obstetrics and Gynecology of Changzheng Hospital Affiliated to Second Military Medical University of Chinese PLA.PARTICIPANTS: Eighty-three healthy women who were hospitalised in Changzheng Hospital and Changhai Hospital affiliated to the Second Military Medical University of Chinese PLA and met the inclusion criteria. Among them, 40 were normal healthy pregnant women(the control group) and 43 were healthy pregnant women with fetal distress(the fetal distress group).METHODS: Radioimmunoassay was used to measure the levels of blood EOP(β-endorphin, dynorphin A1- 13 and leu-enkephalin) of the venous blood of the pregnant women in fetal distress group and the control group and the EOP level in the umbilical blood of the newborns. Also, blood gas analysis of the blood from the umbilical artery was conducted.MAIN OUTCOME MEASURES: The levels of EOP in the venous blood of two groups of pregnant women and the umbilical blood of newborns and the correlation of EOP level with fetal distress.RESULTS: The levels of the umbilical artery blood EOP(β-endorphin,dynorphin A1-13 and leu-enkephalin) in the fetal distress group[(453± 68 ) ng/L, (242 ± 33)ng/L, and(498 ± 68)ng/L respectively] were significantly higher than those in the control group[ (251 ± 39) ng/L, (103± 22 )ng / L and(322 ± 40 )ng / L respectively ( t = 2. 713,2. 762, P< 0.01; t = 2. 132, P < 0.05 ) ]. The umbilical artery blood gas analysis;pH was (7.0 ± 0. 1 ) , PO2 was ( 1.7 ± 0.6) kPa, PCO2 was (8.9 ± 0. 7) kPa.The levels of β-endorphin were negatively correlated with pH and PO2 of the umbilical artery blood(r= -0.418 and -0.437, P < 0.01), but they were positively correlated with PCO2( r = 0. 442, P < 0. 05) . The level of dynorphin A1-13 was negatively correlated with pH and PO2( r = -0. 337,-0.383, P < 0.05), but it was positively correlated with PCO2(r= 0. 346, P < 0. 05). There was no significant difference among the three kinds of blood EOP of the two groups( P > 0.05).CONCLUSION: EOP participates in the pathological progress of the fetal distress and was closely correlated with the occurrence and development of the fetal distress. This finding has a reference value for early rehabilitation and intervention after the fetal was born that can be tested quantitatively.

ABSTRACT

Objective To explore the effects of brain derived neurotrophic factors (BDNF) on concentrations of cerebral endogenous opioid peptides(EOP)in neonatal rats subject to with hypoxic ischemic brain injury (HIBI). Methods Seven day old Sprague Dawley rats were randomly divided into a HIBI+BDNF group (group A),a HIBI group (group B) and a sham operation group (group C). Models of HIBI were established by use of permanent ligation of the left common carotid artery followed by 2.5 hours ofinhalation of 8%O 2+92%N 2, then 0.5 ?g BDNF was injected into the parietal cerebral ventricle in group A immediately. Contents of dynorphin A 1 13 like, ? endorphin like and leu enkaphalin like immunoreactivities (ir DynA 1 13 , ir ? EP and ir LEK) in cortex and hippocampus were measured at 0, 60, 120 min after administration of BDNF. Results The concentrations of ir DynA 1 13 and ir ? EP in the cortex and hippocampus in group B were increased significantly than those in group C at most time points( P

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the postburn change in hypothalamic paraventricular beta-endorphin and the roles of delta-receptor in scalded rats.</p><p><b>METHODS</b>Male Sprague-Dawley (SD) rats were randomly divided into 3 groups, i.e. ICI174864, DPDPE and control groups. The rats were inflicted with 20% TBSA of III degree scalding on the back by boiling (100 degrees ) water. The postburn change in the tissue content of the hypothalamic paraventricular beta-endorphin was determined by radioimmuno assay (RIA). The effects of delta-receptor in scalded shock rats were investigated by observing the change of the rats'survival time and cardiac indices after the micro-injection of delta-receptor agonist DPDPE or antagonist ICI174864 into the hypothalamic paraventricle.</p><p><b>RESULTS</b>(1) The tissue content of the hypothalamic paraventricular beta-endorphin increased significantly (P < 0.01) at 1, 2 and 4 postburn hours (PBHs) in the scalded rats. (2) When compared with that of control group, the ratio of the cardiovascular parameters [mean arterial pressure (MAP), dp/dt(max) and HR] were obviously increased at different time points in rats with pre-injection of ICI174864 whereas the ratio was decreased when DPDPE was used. Nevertheless, the change in the heart rate ratio was not obvious whether ICI174864 or DPDPE was used. (3) The average animal survival time in ICI174864 group was much longer than that in DPDPE group.</p><p><b>CONCLUSION</b>An excessive increase in hypothalamic paraventricular beta-endorphin was one of the factors leading to the aggravation of burn shock and earlier death. delta-receptor located in the tissue might have played important roles in the mediation of the action of hypothalamic paraventricular beta-endorphin. It is beneficial to antagonize the action of delta-receptor for the correction of burn shock and for the prolongation of the of life of animals.</p>

ABSTRACT

To explore the effects of brain derived neurotrophic factor (BDNF) on hypoxic ischemic encephalopathy in neonatal rats,0 2, 0 5 or 2 0?g of BDNF was injected into cerebroventricles of 7d neonatal Sprague Dawley rats with hypoxic ischemic encephalopathy by ligation of left common carotid artery and 2 5h inhalation of humidified 8% O 2 +92% N 2 at 37℃. Its effects on brain edema, levels of malondialehyde (MDA) and neuronal apoptosis at the left cortex and hippocampus were observed. The results showed that brain edema was markedly alleviated in 0 5?g group compared to the vehicle group 24h after BDNF injection. Levels of MDA in the left hemisphere in 2 0?g group were significantly higher than those in either vehicle group or 0 5?g group. Neuronal apoptosis decreased to a different extent in the three groups while obviously in both 0 2?g and 0 5?g groups. It is suggested that BDNF exerts a prominent protective effect on hypoxic ischemic encephalopathy in neonatal rats, but this effect is lowered when overdose is injected.

ABSTRACT

Objective:To investigate the environmental factors a nd individual personalities of anorectic children in order to improve their nu rsing care.Methods:Forty-six children with anorexia caused by emotional disorders and 50 healthy children were tested by a societa l-psychological factor questionnaire and the Eysenck Personality Questionnaire (Junior) to observe their external environmental factors and individual persona lities, respectively. Results:Encidence of pressures from school work, forced eatin g by parents and family discord emerged more frequently in the anorectic group than in the co ntrol group.Scores in the anorectic group were significantly lower in the scale E level and markedly higher in the scale N level than those of the control group (P<0.01).There was no significant difference between the two groups in the P and L scales .Conclusion:The three factors mentioned above were most closely related to onset of disease, among the factors we observed. Both an intro verted personality and unstable emotions are key, characteristics that can induc e anorexia when affected by these factors.Comprehensive nursing care, mainly cons isting of psychiatric-behavioral correction,is proposed to obviate the inducing factors and rectify emotional disorders to promote recovery from anorexia.

ABSTRACT

Objective To investigate the neuroprotection of different brain regions after transplantation of genetically modified myoblasts producing brain derived neurotrophic factor (BDNF) into cortices of neonatal rats subjected to hypoxic ischemic encephalopathy (HIE). Methods Seven day old pups were randomized into sham operated group (C), HIE+BDNF group (B) and HIE+mock transplantation group (A). A rat myoblast cell line expressing and secreting BDNF (BDNF(+)/L 6TG) was constructed. A stereotaxical intracerebroparenchymal transplantation of either BDNF(+)/L 6TG (B) or BDNF(-)/L 6TG (absence of BDNF, A) at 0.8 ?l of cell suspension (4?10 4/?l) into the left cortex of the brain was carried out shortly after HIE undergone by ligation of left common carotid artery followed by a 2.5 h inhalation of humidified 8% O 2+92% N 2 at 37℃. Changes of areas of different brain regions of cortex, hippocampus and striatum at different sections were observed 21 d after the procedure. Injury severity scores of brain tissues were also performed. Results Twenty one days after the manipulation, various degrees of atrophy were observed in the three regions of left hemispheres in group A or B versus contralateral corresponding regions or group C whereas this atrophy in group B was significantly reduced when compared to group A. Not only was the atrophy of cortex near the grafting point alleviated but some other regions of left hemispheres. A similar change pattern was seen in injury severity scores of the tissue. Conclusion Present data suggest that intracerebral transplantation of genetically modified myoblasts producing BDNF has a beneficial effect on the protection from damage of both circumscriptus brain tissues and some other areas distanced from the grafted place.

ABSTRACT

AIM To observe the effects of different interval administration of exogenous brain derived neurotrophic factor (BDNF) on hypoxic ischemic encephalopathy in neonatal rats. METHODS BDNF (0 5 ?g) was microinjected intracerebroventricularly at 0, 1 and 4 h after the hypoxic ischemic encephal opathy in 7 d neonatal Sprague Dawley rats undergone by ligation of left common carotid artery followed by a 2 5 h inhalation of humidified 8% O 2+92% N 2 at 37℃ immediately after the injury, respectively. Changes of brain edema, levels of malondialehyde (MDA) and neuronal apoptosis at the left cortex and hippocampus were investigated 24 h after the injury. RESULTS The most prominent effect of BDNF was seen in 0 h group with a marked decreases in brain edema and levels of MDA and a significantly alleviated neuronal apoptosis while it was lowered obviously when administered at 4 h after the insult. CONCLUSION BDNF exerts a prominent protective effect on hypoxic ischemic encephalopathy in neonatal rats when given as early as possible after the injury rather than later administration.

ABSTRACT

Indexes of cardiovascular function and survival time were obser-vedin rats given the following antagonists respectively after scald injury: anti-p-endorphin serum at 10?l, Naloxone at 2mg, ICI174864 at 0.2mg, or TRH at 2 mg, and half of the doses were administered at 1, 2, 3h after scald. The results showed that the cardiac indexes (dP/dtmax, -dP/dtmax and LVSP)were improved, the decrease of mean arterial pressure ( MAP ) and heart rate ( HR ) were delayed after the injections of anti-p-endorphin serum, naloxone or ICIi748e4, and survival time was significantly prolonged in anti ? - endorphin serum group. TRH had little effect on cardiac indexes, MAP and HR were maintained at high level at earlier period, but sharply sloped down in about 210 min after burn. The result suggests that intraventri-cular administration of anti-?-endorphin, naloxone or ICI174864 had much benefit on scald shock, but TRH was uncertain at least in the treatment of scald shock.
